چکیده

This study aimed to explore the therapeutic efficacy of tripterygium glycosides for experimental autoimmune myasthenia gravis (EAMG). EAMG model was established in rats, and the rats were divided into complete Freund’s adjuvant group, EAMG model group and tripterygium glycosides treatment group. All rats were evaluated by Lennon scoring and body mass monitoring. We found that the treatment with tripterygium glycosides improved the clinical symptoms of myasthenia gravis, reduced Lennon score but increased body mass. Furthermore, tripterygium glycosides effectively reduced the activities of specificity-activated antigen-reactive T cells and the expression and secretion of pro-inflammatory cytokines such as IL-17 and IFN-γ. In conclusion, tripterygium glycosides are a promising agent for the treatment of autoimmune myasthenia gravis.

O 29: Autoimmune Myasthenia Gravis Introduction, Immunopathogenesis and Classification

Autoimmune Myasthenia Gravis (MG) is a unique disease among all autoimmune disorders in two ways. First, there are a wide range of sub-specialties involved in the diagnosis and management of MG; secondly MG is an autoimmune disorder whose autoimmunity is well established. In this paper I will cover these topics: history and epidemiology of MG followed by a brief overview on physiology of neurom...

Suppression of ongoing experimental myasthenia by oral treatment with an acetylcholine receptor recombinant fragment.

Myasthenia gravis (MG) is an autoimmune disorder in which the nicotinic acetylcholine receptor (AChR) is the major autoantigen. In an attempt to develop an antigen-specific therapy for MG, we administered a nonmyasthenogenic recombinant fragment of AChR orally to rats. This fragment, corresponding to the extracellular domain of the human AChR alpha-subunit (Halpha1-205), protected rats from sub...

Association of HLA-DQA1*0101/2 and DQB1*0502 with Myasthenia Gravis in Southern Iranian Patients

Background: Myasthenia gravis is an autoimmune disorder of neuromuscular junction characterized by skeletal muscle weakness and fatigability. Different genes may control the induction and clinical presentation of this disease. Various HLA alleles are reported as predisposing or protective genetic elements in myasthenia gravis. Objective: The aim of this study was to investigate the probable as...

Effect of hypothalamic lesions on experimental autoimmune diseases in rats.

The development of experimental autoimmune encephalomyelitis (EAE) was prevented in rats immunized with encephalitogenic antigen two weeks, but not twelve weeks, after stereotaxic electrolytic destruction of the anterior hypothalamus. Serum antibody level to the antigen myelin basic protein was decreased, and in vitro lymphocyte transformation response to a mitogen was increased. On the other h...

Degradation of acetylcholine receptor in diaphragms of rats with experimental autoimmune myasthenia gravis.

The degradation of acetylcholine receptor observed in denervated and innervated normal rat diaphragms in organ culture is stimulated by exogenous antireceptor serum. In this paper we demonstrate that diaphragms from rats with experimental autoimmune myasthenia gravis contain reduced amounts of acetylcholine receptor. Acetylcholine receptor from myasthenic, but not from normal, rats has antibody...
